Can't load autoexec.nt

I'm trying to load a program (16-bit Windows-based program), and I'm getting an error. ((C:\Windows\System32\Autoexec.nt The system file is not suitable for running MS-DOS and Microsoft Windows applications. Choose 'Close' to terminate the application)). I tried doing what was on the microsoft website to fix it, but it doesn't seem to be working, anyone know how to?
